Ripretinib API

Mechanism of Action

Ripretinib is a small molecule tyrosine kinase inhibitor that targets multiple kinases, including KIT, PDGFRα, and FGFR1. It works by inhibiting the activity of these kinases, which are involved in the regulation of cell growth, proliferation, and survival.

In particular, ripretinib selectively binds to the ATP-binding pocket of KIT and PDGFRα, blocking their activation and downstream signaling pathways. This inhibition leads to a reduction in the growth and proliferation of tumor cells, and ultimately to cell death.

Ripretinib has been approved by the U.S. Food and Drug Administration (FDA) for the treatment of advanced gastrointestinal stromal tumors (GIST) in adults who have received prior treatment with at least three kinase inhibitors, including imatinib, sunitinib, and regorafenib.

Indication

Ripretinib (brand name Qinlock) is a tyrosine kinase inhibitor that has been approved by the U.S. Food and Drug Administration (FDA) for the following indications:

Advanced gastrointestinal stromal tumors (GIST): Ripretinib is indicated for the treatment of adult patients with advanced GIST who have received prior treatment with at least three kinase inhibitors, including imatinib, sunitinib, and regorafenib.

Fourth-line advanced GIST: Ripretinib is also indicated for the treatment of adult patients with fourth-line GIST.

Gastrointestinal stromal tumors (GIST) are a type of cancer that begins in the gastrointestinal tract, usually in the stomach or small intestine. Ripretinib is used to treat advanced GIST that has spread to other parts of the body (metastatic) and is no longer responding to other treatments.
